Peart (shoulder) has been ruled out ahead of Sunday's game against Buffalo, Dan Salomone of the Giants' official site reports.
The 26-year-old tackle suffered a shoulder injury sometime in the last week and will miss his first game of the season this coming Sunday. Peart plays a rotational role on a depleted Giants offensive line that's already without Andrew Thomas (hamstring) and John Michael Schmitz (shoulder).
